Treating the untreatable with a new generation of medicines that truly reprogram the immune system.

Oryn Therapeutics is advancing a novel class of immune-modulating macrocyclic peptides that, rather than broadly inhibiting immune function, address the underlying disease drivers of chronic inflammatory and autoimmune diseases while preserving the body's essential protective responses.

About Oryn

Patients deserve therapies that reach far beyond conventional immune suppression.

For the first time, we can combine evolutionary immune biology and next-generation precision medicine—pioneering Orynotides™, a new platform of macrocyclic peptide therapeutics designed to access challenging extracellular and intracellular biological targets simultaneously, and to precisely modulate dysregulated immune networks using a single agent.

Science & Platform

A macrocyclic peptide platform built to engage the biology others can't.

Orynotides™ occupy the middle ground between small molecules and biologics, compact, constrained, cell-permeable peptides engineered to engage both extracellular and intracellular targets and to reprogram dysregulated immune networks rather than broadly suppress them. Four principles define the platform.

Many of the most important biological drivers of inflammatory, autoimmune, and fibrotic diseases remain inaccessible to conventional therapeutics. While small molecules often lack the structural complexity required to selectively engage these targets, and biologics are generally limited to extracellular interactions, Orynotides™ are engineered to occupy the unique middle ground between these modalities. Their highly constrained macrocyclic architecture enables engagement of complex protein surfaces and allosteric regulatory domains that have historically proven difficult or impossible to modulate therapeutically. As a result, Orynotides have the potential to unlock validated disease pathways that have remained beyond the reach of traditional drug discovery approaches.

Our platform is designed to selectively modulate biological networks rather than simply block a single signaling pathway. By targeting allosteric control points on proteins that are key regulators of inflammatory and tissue-remodeling pathways, Orynotides may enable precise reprogramming of dysregulated immune responses while preserving normal physiological function. This approach has the potential to address the underlying biology driving disease progression across inflammatory, autoimmune, and fibro-inflammatory conditions, creating opportunities to deliver transformative therapies where existing medicines have failed to provide durable disease control.

Most therapies for autoimmune and inflammatory diseases are designed to block a single cytokine, receptor, or signaling pathway. While these approaches can reduce inflammation, they often fail to address the broader biological networks that drive disease progression. As a result, many patients experience incomplete responses, loss of efficacy over time, or continued disease activity despite treatment.

Orynotides™ are built on a fundamentally different principle: immune reprogramming. Rather than shutting down individual components of the immune system, Orynotides are designed to reshape the behavior of interconnected inflammatory networks. By influencing upstream regulatory mechanisms that coordinate multiple downstream pathways simultaneously, a single Orynotide has the potential to drive complex biological systems toward a desired therapeutic state. This network-based approach may enable broader and more durable disease modification than can be achieved through conventional pathway-specific therapies.

The immune system is governed by a dynamic network of extracellular signals, intracellular pathways, and transcriptional programs that collectively determine how disease progresses. Most therapeutic modalities are limited to targets located either outside or inside the cell, restricting their ability to influence these interconnected biological systems. Orynotides™ are uniquely designed to operate across multiple layers of biological regulation, with the potential to simultaneously modulate extracellular signaling networks, intracellular inflammatory pathways, and downstream cellular responses. This capability enables a single therapeutic agent to exert coordinated effects across complex disease-driving networks rather than acting on a single molecular pathway in isolation.

Importantly, each Orynotide can be engineered to produce a distinct pattern of network modulation, allowing different molecules within the platform to generate differentiated biological outcomes. Rather than creating a series of compounds that all behave similarly, we are developing a family of precision-engineered macrocyclic peptides capable of fine-tuning complex biological networks toward specific therapeutic objectives. This programmable approach creates the potential to develop medicines tailored to the unique biology of individual diseases while leveraging a common platform architecture.

Historically, peptide therapeutics have been limited by challenges related to stability, tissue distribution, cellular access, and oral delivery. Orynotides™ were designed from the outset to challenge these limitations. Their compact macrocyclic architecture, structural stability, and rapid cellular uptake create the opportunity to develop peptide medicines with properties traditionally associated with multiple therapeutic modalities.

Beyond any individual product candidate, Oryn is building a platform capable of generating novel medicines against a broad range of biological targets and disease pathways. The same design principles that underpin the Orynotides platform are being applied to create molecules with distinct target profiles, biological activities, and therapeutic objectives across multiple disease areas. As our understanding of macrocyclic design, cellular uptake, and network biology continues to advance, Orynotides will unlock entirely new classes of medicines that combine target versatility, oral delivery, and programmable molecular engineering.

Our Foundation

Programmable biology.

At Oryn, we believe the future of medicine will not be defined by the ability to block a single target, but by the ability to precisely direct complex biological systems toward healthier states.

Disease emerges when intricate networks of proteins, cells, and signaling pathways become dysregulated. The next generation of therapeutics must move beyond pathway inhibition and toward the deliberate programming of biology itself. Orynotides™ carry millions of years of evolutionary optimization for stability and biological activity, and they were created to make this vision possible.

By combining the structural sophistication of macrocyclic peptides with the ability to engage challenging biology across extracellular and intracellular environments, Orynotides have the potential to influence complex disease networks in ways that traditional therapeutics cannot. Each Orynotide can be engineered to generate a distinct biological response, enabling medicines designed not simply to treat disease, but to direct biology toward specific therapeutic outcomes.

We envision a future where medicines are no longer limited by the constraints of conventional drug modalities, where programmable macrocyclic therapeutics can be designed to selectively engage previously inaccessible targets, reshape disease-driving networks, and create entirely new therapeutic possibilities across human disease. Through Orynotides™, Oryn is building the foundation for a new era of programmable biology, where the ability to precisely engineer biological outcomes becomes one of the most powerful tools in medicine.
